Advanced Flow Cytometry UnitRoom 1.24, Ext. 3102 General InformationThe mission of the Advanced Flow Cytometry Unit (AFCU) in IBMC&INEB is to offer researchers efficient and reliable flow cytometric services with the highest standards of quality control and productivity.
The AFCU provides investigators with equipment and support for acquisition, analysis of flow cytometric data and for cell sorting (separation) from single cell suspension using fluorescence.
For analysis purpose, the AFCU is equipped with benchtop alignment-free multipurpose analyzers from BD Immunocytometry Systems (FACS Calibur and FACSort). The laser configurations on both FACS Calibur and FACSort instruments allow for analysis of cell samples stained with 488 nm excited dyes. Meanwhile, the FACS Calibur equipped with the 635 nm laser for additional FL4 analysis. A number of applications, including the multicolor analysis of cell phenotype, gene expression, membrance potent and cell cycle could be performed. People can make reservation online. The analyzers are available for use directly by the investigators after proper training.
For cell sorting, the AFCU is equipped with a FACS Aria sorter (with 488 nm and 633 nm laser). This instrument provides high-speed, high purity (up to 99.0%) multilaser cell sorting. It provides sterile sorts of single cells directly into tubes (BD Falcon), into various plates as single or multiple cells/well or onto microscope slides for analysis. Availiable FacilitiesFACS CaliburThe benchtop FACS Calibur has been in operation since 2000. It is equiped with a 488 nm blue laser and a 635 nm red laser. It has 4 fluorescence detectors plus forward and sider scatter detectors. (2 Euro/hour for internal user).
Note:PE_Cy5 should not be combined with any FL4 fluorochrome FACSortThe FACSort was installed in IBMC in 1993. It is equiped with a 488 nm blue laser. It has 3 fluorescence detectors, plus forward and sider scatter detectors. (2 Euro/hour for internal user).
FACS AriaThe FACS Aria is a benchtop high-speed sorter, in use since December 2005. It is equiped with 2 lasers: 488 nm and 633 nm. Cells can be sorted at the rate of 20K/sec, with 99+% purity, using a variety of commonly used fluorochromes. Cells can be collected into 5 or 15 ml tubes or into various microplates, as single or multi cells/well or onto microscope slides. (2 Euro/hour for internal user).
