Recombinant protein expression in prokaryotes and yeast

The recombinant protein expression in prokariotes and yeast facility is composed of all that is needed to do expression of recombinant proteins in bacteria and yeast cells ranging from molecular biology tools (vectors, host strains, etc) and cell growth. Equipment in this resource includes Refrigerated incubators shakers: Twin Minitron Incubator Shakers (INFORS AG)

 

Recombinant protein expression in insect cells

The insect cell recombinant protein expression facility is composed of all that is needed to do expression of recombinant proteins in insect cells using baculovirus based vectors, such as: molecular biology tools, cell growth and cell manipulation. Equipment in this resource includes:

  1. shakers (Certomat RM, Sartorius Stedim),
  2. laminar flow  hood (MSC-Advance, Thermo Scientific)
  3. microscope (Eclipse TS100, Nikon)

 

 

Protein purification

The protein purification facility is composed of all that is needed to do purification of recombinant proteins and proteins from natural sources, such as: cell harvesting, cell lysis, chromatography, filtration, buffer exchange, sample concentration and freeze drying.

Equipment in this resource includes:

  1. High capacity (6L) centrifuge Avanti J26XPI (Beckman Coulter),
  2. High-pressure homogenizer French® press (Thermo IEC),
  3. sonifier Branson 250,
  4. tangential filtration Vivaflow 200 (Sartorius) and Pellicon 2 (Millipore),
  5. ultrafiltration Amicon stirred cells (Millipore) ,
  6. 4 preparative liquid chromatography systems (BioLogic LP and Biologic Duoflow from Bio-Rad; AKTAprime plus and AKTA Purifier 10 from GE Healthcare),
  7. an analytical HPLC (L7000 series, Merk-Hitachi) and
  8. a freeze dryer Bentch top pro, 9L, -85ºC (Virtis).

  • Spectroflurometer, photon counting with polarizers and stirred cell, For steady state and anisotropy measurements., Fluoromax-4 (Horiba)

 

  •   Microplate Spectrofluorometer, for steady state and time-resolved fluorescence and luminescence measurments, Molecular devices SPECTRAmax GEMINI XS  

  • Circular Dichroism Spectrometer, For structural characterization of Biomolecules, J815 (Jasco)    

  • SPR Biosensor, For the kinetic characterization of interactions, Biacore X100 2.0 (GE Healthcare)    

  • Isothermal Titration Calorimeter, For the thermodynamic characterization of interactions, Microcal VP-ITC (Malvern)    

  • Double beam UV-Vis spectrophotometer with thermostatic control and 6 cell positioner, Shimadzu UV-2401 PC (Shimadzu Corporation)
